МОЛЕКУЛЯРНА ДІАГНОСТИКА YERSINIA RUCKERI

Мета. Проаналізувати нуклеотидні послідовності гену 16S рРНК вірулентних штамів Yersinia ruckeri та розробити метод молекулярної експрес-діагностики збудника єрсініозу.

Методика. За допомогою алгоритму ClustalW у програмному забезпеченні MEGA версії 6.0 було проаналізовано нуклеотидні послідовності гену 16S рРНК вірулентних штамів Y. ruckeri. Для розроблення молекулярної експрес-діагностики Y. ruckeri використовували метод полімеразної ланцюгової реакції (ПЛР). Підбір олігонуклеотидних праймерів здійснювали у програмному забезпеченні VectorNTI11 та онлайн-сервісі BLAST. Продукти ампліфікації були досліджені за допомогою методів секвенування та аналізу нуклеотидних послідовностей.

Результати. На основі ПЛР розроблено метод молекулярної експрес-діагностики збудника єрсиніозу, бактерії Y. ruckeri. Показано, що специфічні олігонуклеотидні праймери продукують амплікони розміром 600 пар нуклетидів. Продукти ПЛР досліджено секвенуванням та показано, що праймери фланкують ділянку мішені ампліфікації.

Наукова новизна. В межах висококонсервативного гена 16S рРНК бактерій Yersinia виділено локус ДНК, до якого підібрано олігонуклеотидні праймери для експрес-діагностики вірулентного штаму Y. ruckeri.

Практична значимість. Експрес-діагностика єрсиніозу дозволить ідентифікувати збудника цього інфекційного захворювання, бактерію Y. ruckeri, та здійснювати профілактичні або лікувальні заходи в рибогосподарських підприємствах України.
